This article describes a simple chromatographic technique that has been developed and validated for the quantitative analysis of total FOS content during fermentation. The authors used high-performance liquid chromatography (HPLC) coupled to a differential refractive index detector to monitor FOS production during sucrose fermentation by Aspergillus aculeatus. This method applies to both laboratory and industrial settings, allowing tracking of large-scale FOS production and control of the main substrate (sucrose) as well as secondary byproducts such as glucose and fructose.
