This article describes an HPLC-MS/MS method for quantitatively analyzing eight key neurochemicals in brain tissue. The primary application of this method was to investigate alterations in neurotransmitter profiles in a rat model of Alzheimer's disease (AD). The study meticulously compared the concentrations of these neurochemicals across three distinct groups: healthy control rodents, those afflicted with induced AD, and AD-affected rats that received therapeutic interventions, specifically an extract from Xanthoceras sorbifolia or huperzine. The investigation revealed significant imbalances in neurotransmitter levels in the AD model. Notably, dopamine, norepinephrine, and acetylcholine concentrations were observed to decline, while L-tryptophan levels exhibited an increase. Intriguingly, the administration of X. sorbifolia extract and huperzine demonstrated a restorative effect, bringing the levels of acetylcholine and L-tryptophan back toward a more typical range. These findings suggest that acetylcholine and L-tryptophan could potentially serve as indicators for monitoring the effectiveness of treatments for Alzheimer's disease.
