Comprehensive Glycoprofiling of Tumor Cell Line at Creative Biolabs

Glycoprofiling of tumor cell lines mainly involves the following steps:

  • Culture of tumor cell lines: We culture tumor cell lines under sterile conditions to ensure good cell growth.
  • Cell collection: We collect tumor cells using appropriate methods (such as trypsinization).
  • Washing of cells: We wash the cells with a buffer such as saline or PBS to remove impurities produced in the culture medium and during cell culture.
  • Cell lysis: We use an appropriate cell lysis buffer to lyse the cells and release the internal components.
  • Precipitation of proteins: We centrifuge the cell lysate and collect the supernatant. Then add solvents such as ethanol or acetone to precipitate the protein to remove proteins that will affect subsequent analysis.
  • Extraction of sugars: We use a mixed solvent system such as chloroform-methanol to extract sugars and separate the organic layer and aqueous layer. The organic layer is then transferred to a new tube and the solvent is removed by concentration and drying.
  • Derivatization: We perform derivatization on sugar samples, such as methylation, etc. to facilitate subsequent analysis.
  • Glycoprofiling of tumor cell line: We dissolve derivatized sugar samples in appropriate solvents to facilitate further chromatography or mass spectrometry analysis. For prepared tumor cell line samples, we use a variety of techniques to perform glycoprofiling:
    • Chromatography technology: We inject the sample into the chromatography system and separate the different sugar components through column separation technology. Subsequently, appropriate detectors are used for detection and quantification, resulting in a glycan profile.
    • Mass spectrometry: We perform glycan analysis through a variety of mass spectrometry techniques, which are suitable for analyzing sugars in complex mixtures and have high sensitivity and accuracy.
    • Nuclear magnetic resonance (NMR): It is used to determine the structure of sugars, identify isomers, and perform quantitative analysis.
    • Fluorescent labeling technology: We label sugars with specific fluorescent labeling agents and perform qualitative and quantitative analysis through fluorescence detection technology.

Results: Cisplatin is a chemotherapeutic drug for the treatment of testicular cancer (TC). The authors analyzed the glycan changes of four different TC cell lines and their cisplatin-resistant sublines through the ELLA protocol. They chose the TC biomarker human chorionic gonadotropin (hCG) as the target for glycoprofiling analysis. hCG is present in the cytoplasm of cell lysates, and the authors immobilized the antibody on an ELISA plate and incubated it with the cytoplasm of the cell lysates and a variety of lectins. Results showed that dolichos biflorus agglutinin (DBA) differentiated between resistant (shown by red bars) and sensitive TC cell lines (shown by green bars).
