This research details the development and validation of a sophisticated UHPLC-MS/MS analysis technique, designed to precisely measure AA and its various metabolic byproducts. These specific metabolites are generated through the cyclooxygenase (COX) and lipoxygenase (LOX) pathways within biological tissues. The study's primary application of this method was to investigate changes in these compounds in rat models of benign prostatic hyperplasia (BPH). The findings revealed a significant elevation in the levels of eleven distinct biomarkers across multiple tissues, including the prostate, seminal vesicles, bladder, liver, and kidneys, in the BPH-affected rats when compared to a control group. This suggests that BPH intensifies the production of arachidonic acid and its eicosanoid derivatives, likely by promoting the activity of COX and LOX enzymes.
