B Cell based Inhibition by Antigen Assay Service

Creative Biolabs' B cell-based inhibition by antigen assay delivers a clear, actionable solution for your antibody discovery program. The key problem we solve is the critical gap between simple binding affinity and true functional efficacy. Creative Biolabs provides a robust platform to confirm that your candidate antibodies not only bind to the target antigen but also effectively block its biological function on a native cell surface. This ensures that you advance only the most promising candidates to the next stage, saving significant time and resources.

The Valuable Inhibition Assay for Epitope Identification

One of the most important mechanisms of action of antibodies for therapies is the antibody-based inhibition function, such as inhibiting key signaling transduction for tumor cell growth. In prostate cancer, antibodies are developed to block the prostate-specific antigen (PSA) binding to a1-antichymotrypsin (ACT). Immune checkpoint antibodies are developed to inhibit the interaction of PD-1 on T cells with PD-L1 on tumor cells. The binding interaction between antibody and epitope peptide inhibits the binding of antigens of homologous origin to the same antibody. This antigen inhibition assay has become a classical method for identifying antigenic epitopes within antigens.

FAQs

What is the primary advantage of this assay compared to traditional ELISA or SPR?

Our assay uses native B cells, which present the antigen in its natural, membrane-bound form. This provides a more physiologically relevant context than purified proteins in a well plate, allowing us to screen for true functional inhibitors and reduce false positives.

How does this assay help with epitope mapping?

While our primary goal is functional inhibition, the assay can provide valuable insights into epitope relevance. By demonstrating that an antibody can block antigen binding on a native cell, we confirm its ability to bind to an accessible and functionally relevant epitope, a crucial step that complements and validates peptide-based epitope mapping.
