Traditional organic sonosensitizers often yield poor therapeutic effects due to their inherent hydrophobicity. This research presents a groundbreaking approach: leveraging sonosensitizer aggregation to significantly boost their sonodynamic activity. The study focused on phthalocyanine–artesunate couplets, revealing that their aggregated forms dramatically increased ROS generation. Notably, ZnPcT4A showed a remarkable 60-fold ROS production increase when aggregated in water, a phenomenon also observed in other sonosensitizers, suggesting a universal aggregation-enhanced sonodynamic activity. This discovery fundamentally challenges the conventional avoidance of aggregation in sonosensitizer design, opening a new avenue for developing highly efficient organic sonosensitizers. Both in vitro and in vivo studies confirmed ZnPcT4A aggregates' high biosafety and potent anticancer efficacy, achieving 98% tumor growth inhibition. This research marks a significant step forward in sonodynamic therapy.
