Peptide-MHC Tetramer/Multimer
The exquisite specificity of T cell response to peptide/MHC is useful in tracing antigen-specific T cells and analyzing antigenic peptides. But the binding affinity of a single peptide/MHC to its cognate TCR is low and dissociates quickly. Tetramerized MHC molecules loaded with candidate peptides promoted the association of TCR with the complex. When labeling with fluorescent dyes, MHC tetramer/multimer could act as a staining reagent for epitope-specific T cells, enabling the convenient detection by a flow cytometer. Class I and class II tetramer reagents are produced differently and have been used in identifying CD8+ and CD4+ T cells, respectively.
