Introduction of Chromatography Assay
Affinity-based chromatography is a powerful and specific separation method used for bioactive compound isolation and purification employing the binding affinity between immune complexes. Specific monoclonal antibodies or patient immunoglobulin have been used to capture human or mouse tumor tissue-derived antigens, HLA-restricted tumor-specific antigens, and neoantigens. Following the capture process, the proteins bound to the chromatography are eluted and evaluated by various other methods.

FAQs
What are the key advantages of this method for discovering antibodies from immunized animals?
The primary advantage is speed and specificity. Our assay significantly compresses the timeline from immunization to candidate identification. By directly selecting for antigen-specific B cells, we ensure that the candidates you receive have already undergone natural affinity maturation.
Can this assay be used to identify antibody fragments or scFv?
Yes. We can isolate B cells that produce a variety of antibody formats, including fragments. As demonstrated in published research, specialized affinity ligands such as Protein L can be effectively used for purifying these fragments, and our upstream assay is perfectly compatible with these downstream purification protocols.
