Publication
This publication introduces a rapid-acting therapeutic for peanut allergy, addressing limitations of current treatments. The research aimed to develop a treatment using allergen-specific IgG4 antibodies, guided by human IgE antibodies from allergic individuals. Researchers used single-cell RNA-sequencing on blood samples to identify IgE mAbs, which were then class-switched to IgG4. These recombinant IgG4 mAbs were evaluated for specificity, affinity, and activity using various in vitro and in vivo tests, including a peanut allergy mouse model. Results showed that human peanut-specific IgE mAbs target key epitopes on Ara h 2 and Ara h 6. The recombinant IgG4 mAbs effectively blocked these epitopes. Notably, IGNX001, a blend of two high-affinity IgG4 mAbs, offered strong protection against peanut-induced mast cell activation in vitro and anaphylaxis in mice. In conclusion, the study successfully developed IGNX001, a peanut-specific IgG4 antibody therapeutic, demonstrating compelling preclinical efficacy. This therapeutic, derived from human IgE mAbs, warrants further clinical investigation and suggests a promising development strategy for other allergies.
