Studies have shown that many bacterial polysaccharide antibodies provide protection against bacterial infections. Polysaccharides are T-cell independent antigens, and neonates and infants cannot produce antibody responses to such antigens. Therefore, proteins containing T cell epitopes need to be coupled to such antigens to compensate for their inability to induce antibody responses in infants and young children. This conjugation of haptens and carriers is a key step in the development of hapten vaccines and also an important step for hapten vaccines to exert efficacy. The appropriate hapten-carrier conjugation method can only be selected after the functional group on the hapten and carrier be determined. There are generally two types of methods, the spontaneous chemical reaction and the intermediate molecules cross-linkage. The former is suitable for chemically reactive haptens, while the latter is suitable for non-reactive haptens.

CDAP Conjugation Method

Many important polysaccharides require activation and derivatization before binding to proteins. Cyanylation with CNBr is a commonly used method for preparing protein-polysaccharide conjugate vaccines. Although this method is very effective, the toxicity and poor operability of CNBr limit the application of this method. I-cyano-4dimethylaminopyridinium tetra-fluoroborate (CDAP) is a water-soluble cyanylating reagent, and it enables the cyanylation reaction to be carried out under relatively mild conditions. CDAP activates a wide range of acidic, neutral, and other types of polysaccharides that allow these polysaccharides to react with diamines, diterpenes, or other functional groups to prepare for protein conjugation. The advantage of the CDAP conjugation method is that the polysaccharides activated by CDAP can be coupled to proteins without the need for additional molecules, thus simplifying the preparation of hapten-coupled vaccines.

The CDAP conjugation method has been used in a variety of pneumococcal conjugate vaccines and meningococcal conjugate vaccines. In addition, the method is being used in many vaccines such as for pneumococcal and meningococcal diseases, which are currently in clinical trials.
