Marburg virus disease (MVD) is caused by a hemorrhagic fever virus of the Filoviridae family. It is a severe and highly fatal disease that produces symptoms of fever, chills, headaches, and muscle aches early in the disease; symptoms worsen and may lead to hemorrhagic fever and death. MVD is endemic in Africa with case fatality rates of up to 90% and can be transmitted from person to person by exposure to blood and other bodily secretions. Marburg viruses are filamentous particles that may appear in the shape of a shepherd's crook or in the shape of a "U" or a "6", and they may be coiled, toroid, or branched. Actual treatment of the virus after infection is not possible but early, professional treatment like vaccination considerably an effective administration.

Therefore, the multiagent DNA vaccine has the potential of these four agents and point to a possible method of rapid development of multiagent vaccines for preventing naturally occurring or deliberately introduced diseases.

Recombinant Viral Vectors Vaccine

An RNA replicon which was based upon Venezuelan equine encephalitis (VEE) virus was used as a vaccine vector. In the design of the vaccine, the VEE structural genes were replaced by genes for Marburg virus glycoprotein, nucleoprotein, VP40, VP35, VP30, or VP24. Immunization with either killed Marburg virus or a glycoprotein subunit could prevent lethal infection in guinea pigs. As a more definitive test of vaccine efficacy, nonhuman primates were inoculated with VEE replicons expressing Marburg virus glycoprotein and/or nucleoprotein. The experimental group vaccinated with replicons that expressed glycoprotein or nucleoprotein was completely protected from MVD. It gives a choice to develop the vaccines against MVD.

Virus-like Particle (VLP)-based Vaccine

Virus-like particle (VLP)-based vaccines obtain a lot of focus due to the advantage of being morphologically and antigenically similar to the live virus from which they are derived. In the development of Marburg virus vaccine, the glycoprotein and VP40 matrix protein from Lake Victoria marburgvirus (MARV) were spontaneously expressed to product of VLPs in mammalian cells. Animals like Guinea pigs or macaques were vaccinated with Marburg virus VLPs (mVLPs), both types of animals develop humoral and T-cell responses and were completely protected from a lethal homologous MARV challenge, showed the mVLPs could broadly protect against MVD. Thus, mVLPs can be a suitable candidate with the broad immunity and efficient protection against Marburg virus.
