Type 5 adenovirus, belonging to species C adenoviruses, is a linear double-stranded DNA virus with a diameter of about 80-120 nm. It can rapidly infect mitotic and non-mitotic cells. After binding to cell surface receptors, it enters the cells through endocytosis, gathers near the nuclear pore, and releases the DNA of adenovirus into the nucleus after infection and starts protein expression. By modifying wild adenovirus, preserving the packaged signal sequence and removing the coding sequence of the viral protein, it can not only increase the loading capacity of foreign genes but also reduce the cytotoxicity and immune response of the body, thus obtaining a more secure gene import system.

Adenovirus type 5 causes primarily infections of the upper respiratory tract infection and pneumonia. It is also responsible for some cases of conjunctivitis, cystitis, and gastroenteritis. Adenovirus type 5 related diseases are usually diagnosed by the identification of polymerase chain reaction (PCR) or histopathological viral contents, combined with clinical features and viral DNA detection. Research shows that virus detection itself at multiple sites and increasing of viral load are considered to be high-risk.
