Human Astrovirus

In 2008, the first human astrovirus (HAstVs) was found in a sample of pediatric stools in Australia. Human astroviruses have been widely recognized as the leading pathogen of diarrhea caused by virus in children, the elderly, and immunocompromised patients. In addition, there is evidence that HAstVs may be associated with neurological diseases such as encephalitis and systemic infections. HAstVs was identified as three phylogenetic branches, namely canonical HAstVs-1 to 8, containing 8 genotypes, noncanonical genotypes MLB1 to 3 and VA1 to 5. Among them, HAstVs-1 is the most widely prevalent serotype.

Immune Responses to Human Astrovirus

Based on the age-related characteristics of AstV infection in most species, scientists believe that the immune response may provide some protection for the infection of the host by the virus. Clinical studies and epidemiological studies have found that humoral immune responses play a role in limiting viral infections and preventing disease progression. At the same time, the seroprevalence test also showed that most healthy adults possess antibodies against HAstVs. The immune effector molecule after gastrointestinal infection by viruses is mainly mucosal IgA. And the presence of IgA is also detected after AstV infection, however, whether the level of IgA is related to immune protection needs further study. In addition to reflecting clues that humoral immune responses may be involved in combating re-infection, there are also studies from healthy human tissue examinations that found the existing of HAstVs-specific CD4+ and CD8+ T cells. Therefore, in general, healthy people exert anti-secondary infection effects through both humoral and cellular immune responses. Moreover, the restriction of adaptive immune responses to replication of AstV in primary infection was also observed in the mouse model. In addition to the role of adaptive immunity in protecting and controlling viral infections, innate immunity also performs well in limiting and controlling the infection of young people with the virus. In terms of the complement system, it has been reported that the purified recombinant HAstVs capsid protein is capable of inhibiting the complement-mediated hemolysis reaction.

Development of Vaccines against HAstV

There is currently no HAstV vaccine for commercial use. The disease caused by astrovirus is clinically relatively mild, so the ideal strategy for HAstV vaccine is to prepare a multivalent vaccine that can prevent multiple serotypes or different strains. In addition, VLPs of HAstV have been successfully constructed using various expression systems, and their efficacy in preventing viral infections requires further evaluation. Designing a multivalent vaccine using the HAstV viral capsid protein as a target combined with other diarrhea-causing pathogen antigens is a prospect for HAstV preparation.
