Introduction
Synthetically derived monophosphoryl lipid A (MPLAs) from E. coli is a chemically synthesized, pure compound. This compound activates TLR4 but does not trigger TLR2, highlighting its high level of purity. Structurally, it closely resembles natural MPLA, with the key difference being that it contains six fatty acyl groups, whereas bacterial-purified MPLA typically consists of a mixture of lipid A molecules with five, six, or seven acyl chains.
