The detection principle of ELISA kits is to use enzyme-labeled antibodies or antigens as probes, through the specific binding of antigens and antibodies, and then through a series of reactions catalyzed by enzymes to display measurable signals. The main steps are as follows:
Coating: Fix specific antigens or antibodies on microplates.
Add sample: Add the sample to be tested (such as serum, plasma, cell culture supernatant, etc.) to the microplate, and the antibody or antigen in the sample reacts with the antigen or antibody bound to the plate. In this step, the antigen and antibody form a complex.
Washing: Remove unbound components by washing them to reduce interference.
Secondary antibody incubation: Add enzyme-labeled secondary antibody for incubation.
Substrate reaction: Add substrate solution, and the enzyme catalyzes the substrate reaction to produce a detectable colored product. Tetramethylbenzidine (TMB) is usually used as a substrate.
Measurement: After the enzyme reaction is completed, the signal intensity of the colored product is measured at a specific wavelength and calculated according to the standard curve. The signal is usually proportional to the amount of antigen or antibody in the sample.
