B4GAT1 knockout HepG2 cell line lacks B4GAT1 protein, beta-1,4-glucuronyltransferase 1, essential for poly-N-acetyllactosamine synthesis (blood group i antigen determinant) in liver cells. B4GAT1 knockout in HepG2 cells enables study of poly-N-acetyllactosamine synthesis disruption in liver cancer, potentially affecting blood group i antigen expression on hepatocytes and poly-N-acetyllactosamine dependent functions in the liver tumor context, potentially mirroring B3GNT2/3/4 knockouts.
