L-Leucine for SILAC is the standard, unlabeled form of the essential amino acid L-leucine, used as a control or "light" component in stable isotope labeling by amino acids in cell culture (SILAC) experiments. While SILAC relies on comparing cells grown in media containing isotopically labeled amino acids, a baseline is essential. This unlabeled L-leucine is incorporated into the proteins of cells grown in standard media, establishing the "light" proteome. This light proteome acts as the reference against which the "heavy" (labeled) proteome from cells grown in media containing stable isotope-labeled leucine is compared using mass spectrometry. This comparison enables researchers to quantify relative changes in protein abundance between different experimental conditions.
