Description
GlycoFlux™ Lenti-PYGL Viral Particle provides PYGL, the liver glycogen phosphorylase converting glycogen to glucose-1-phosphate; phosphorylation at Ser15 activates the enzyme, and mutations cause GSD VI (Hers disease). This replication-defective vector is intended for generating stable mammalian cell lines for hepatic glucose-output modeling, hormonal/allosteric control assays, and variant rescue with routine selection or fluorescence enrichment.

Transduction Evaluation

Insert Identity Confirmation
All lentiviral vectors at Creative Biolabs are verified for correct proviral integration by PCR. In this assay, cells are transduced with serial dilutions of the lentiviral vector, harvested after a few days, and genomic DNA is isolated. A defined region of the expected lentiviral insert is then PCR-amplified to confirm identity.

Summary
The product of this gene is a homodimeric enzyme that releases glucose-1-phosphate from liver glycogen by cleaving alpha-1,4-glucosidic bonds. Conversion from inactive phosphorylase B to active phosphorylase A occurs via phosphorylation at serine 15. Additional regulation is mediated by allosteric effectors and hormones. Mutations cause glycogen storage disease type VI (Hers disease), marked by moderate hypoglycemia, mild ketosis, growth delay, and hepatomegaly. Multiple isoforms are generated by alternative splicing.
