| Description | GlycoErase™ ACLY knockout HEK293 cell line is a human embryonic kidney (HEK293) model in which ACLY expression has been eliminated. ATP citrate lyase (ACLY) is the primary enzyme generating cytosolic acetyl-CoA from citrate, linking cellular energy metabolism to biosynthetic pathways such as lipogenesis and cholesterol production. Loss of ACLY disrupts acetyl-CoA supply, reshaping lipid metabolism and membrane biogenesis in a human context. The knockout of ACLY in the HEK293 background provides a relevant system for studying metabolic rewiring in cancer cells, testing inhibitors of acetyl-CoA-dependent processes, and exploring links between lipid metabolism and gene regulation. |

| Summary | ATP citrate lyase is a tetrameric enzyme (~440 kDa) that generates cytosolic acetyl-CoA from citrate and CoA, with ATP hydrolysis to ADP and phosphate. Acetyl-CoA supports lipid and cholesterol synthesis, and may contribute to acetylcholine production in neural tissue. Multiple transcript variants encode distinct isoforms. |
