GlycoFlux™ human SIGLEC12 overexpression HepG2 cell line is a stable glycoengineered cell line designed to express SIGLEC12, a member of the SIGLEC3-like subfamily characterized by an extracellular V-set Ig-like domain, two C2-set Ig-like domains, and cytoplasmic ITIM and SLAM-like motifs. Upon tyrosine phosphorylation, SIGLEC12 recruits SHP1/2 phosphatases to mediate inhibitory signaling in macrophages, suppressing immune activation. The gene is located within a SIGLEC cluster at 19q13.4 and generates multiple isoforms through alternative splicing. This system provides a robust model to study sialic acid-dependent inhibitory signaling, tumor immune evasion, and glycosylation-related modulation of innate immunity.
