The O-glycoprotein deglycosylation kit (orela reaction) uses the Orela reagent to chemically react with the terminal monosaccharide on the hydroxyl derivative (usually serine or threonine) in the glycoprotein, thereby breaking the covalent bond between the glycan and the peptide backbone. In this process, the glycan is released in the form of a hydroxyl derivative to form a glycan with a free reducing end. This released glycan is fluorescently labeled in subsequent HPLC and mass spectrometry analysis, thereby achieving relative quantification of different glycan types. One kit contains deglycosylation of 12 glycoprotein samples.
