Creative Biolabs is at the forefront of ATP-responsive liposome technology, we can unlock the full potential of ATP-responsive liposome in the treatment of various diseases.
Adenosine-5'-triphosphate (ATP) is a crucial biomolecule in cellular energy metabolism and signal transduction. A significant ATP concentration gradient exists between intracellular (1-10 mM) and extracellular (<0.4 mM) environments, which is considerably steeper than the pH gradient. This gradient is found in almost all cell types, yet tumor microenvironment may exhibit even higher ATP levels. Thus, a stimuli-responsive drug delivery system that capitalizes on the ATP concentration gradient can facilitate targeted cancer therapy.
Utilizing ATP as a stimulus offers several advantages:
ATP-responsive liposomes represent a novel class of stimuli-responsive liposomes that undergo structural changes upon binding to ATP, leading to the rupture of the lipid membrane and the release of encapsulated drugs. To confer ATP responsiveness to liposomes, functional modules that can distinguish ATP from other cytoplasmic components are required, which include the following two categories:
These are chosen from a vast array of random ssDNA sequences for their high binding affinity to adenosine/ATP in vitro. With their relatively short sequences (around 30 bases), these aptamers are easy to functionalize and exhibit specific reactions, making them a popular choice for ATP-responsive liposome.
ATP-responsive liposomes are a novel class of liposomal delivery vehicles that are designed to undergo conformational changes upon binding to ATP, leading to the disruption of membrane packing and the subsequent release of encapsulated drugs.

Lipid switches are incorporated into the liposome formulation, designed to undergo significant conformational changes upon ATP binding, leading to structural degradation and the selective release of encapsulated drugs.
