As a practical method to reduce off-target toxicity in cancer treatment, prodrugs have attracted widespread attention. Nowadays, nano-drug delivery systems and prodrug strategies have been applied to address this challenge. Prodrugs (drugs conjugated to pro-carriers) are inactive compounds that can be degraded by chemical or enzymatic reactions into parent biologically active drugs in vivo. Prodrugs are used to optimize the functions of the parent drug, such as physicochemical, ADME (absorption, distribution, metabolism, excretion) and pharmacological properties. Prodrug methods can promote preparation and administration, improve drug efficacy, improve site-specificity and/or reduce toxicity. Today, prodrug methods are used in the early stages of drug development. Lipid prodrug approach is an evolving field in improving many drug properties/delivery/therapies, which can provide solutions for various unmet needs.

Liposomal prodrug formulations have several significant advantages, including high drug loading efficiency, improved drug availability, drug release controllability, and less aggregation tendency after encapsulation.
Compared to other types of liposomal drugs, prodrug-based liposomes can respond intelligently to certain stimuli and release the desired drugs. After decades of development, a variety of prodrug-based liposomes with different chemical modifications have emerged as corresponding stimuli, including internal tumor microenvironment (TME) stimuli (such as pH, redox environment, enzyme and hypoxic, etc.) and external environmental stimuli (for example, light, heat, ultrasound, magnetic field, etc.). Most importantly, choosing the appropriate stimulus type and chemical modification in different situations is the key to designing satisfactory prodrug-based nanomedicine.

Liposome encapsulated prodrugs are a special kind of nano-assembled prodrugs, which mainly use liposomes to load or encapsulate anticancer prodrugs through non-covalent binding.
