Background of Liposome-Based Adjuvant

Adjuvants in immunology are usually used to modify or enhance the effects of vaccines by stimulating the immune system to respond more strongly to vaccines, thereby improving immunity to specific diseases. Adjuvants are generally defined as functional excipients and constitute a heterogeneous class of compounds, which can be broadly classified into delivery systems or immunostimulators, many adjuvants have these two properties. Particulate drug delivery system provides the ability to incorporate antigen and immunostimulator within pathogen-sized particles, thus protecting the antigen from degradation and facilitating delivery to antigen-presenting cells. Among the available particulate drug delivery systems, liposomes are the first to be described as an adjuvant system, and their immune and adjuvant properties have been identified.

Liposome-Based Adjuvant Development

In the development of liposomal adjuvant, the type of immune response is related to its physical and chemical characteristics. The size and charge of liposome particles affect the biological distribution of liposomes, the exposure of lymph nodes and the supplement of the innate immune system. Liposome composition and preparation can be selected to achieve the desired characteristics, such as lipid, charge, size, size distribution, antigen or adjuvant encapsulation and location selection. Co-formulations containing different types of antigens or adjuvants can be combined with the above parameters to adjust the individualized application of liposome vaccines.
