LacNAc

Poly-LacNAc (Poly-N-acetyllactosamine) comprises repeated Galβ1-4GlcNAc disaccharides, called N-acetyllactosamine (LacNAc). The biosynthesis of poly-LacNAc is accomplished by the enzymatic action of β-1,3 N-acetylglucosaminyltransferase (β3gnt) which catalyzes the addition of GlcNAc to N-glycan Gal termini. Among the several homologs of β3gnt, β3gnt2 is crucial enzyme for synthesis of poly-LacNAc in CHO cells. Poly-LacNAc structures occur in mammalian glycoproteins in both N- and O-linked glycans. They represent a backbone for additional modifications by sialyltransferases, fucosyltransferases, and sulfotransferases. Poly-LacNAc could serve as a potential age-specific ligand for attachment and infection of Rotavirus P in neonates and infants.

Biological Significance of Poly-LacNAc

  • Cancer Metastasis

Poly-LacNAc in tumor growth and metastasis is the most attractive aspect regarding its biological significance. One leading mechanism is that the N-linked poly-LacNAc glycans are supposed to provide a backbone for attachment to adhesion molecules, which trigger the signaling pathways for tumor cell growth and invasion.

  • Immune Cell Signaling

TCR complex at the cell surface was associated with galectins, which are known to modulate T-cell proliferation and apoptosis. The association is related to β1, 6 branched poly-LacNAc on N-glycans.

  • Receptor for Pathogens

It is known that oligosaccharides serve as specific receptors for bacteria, viruses, and parasites. Some of these glycoconjugates contain poly-LacNAc sequences.

  • Maintenance of Membrane Stability

The decoration of erythrocyte membrane anion transporter by poly-LacNAc is important for membrane stability. The lack of poly-LacNAc leads to aggregation of the membrane glycoproteins and abnormal membrane structures.

  • Intracellular Sorting

In the study on the protozoan parasite, Trypanosoma brucei, the N-linked glycans containing linear poly-LacNAc were recognized as sorting signals for membrane proteins in endocytosis.
