Background: The Diversity of Bacterial Surface Glycans

The surface of bacteria contains numerous complex and varied carbohydrate structures. These surface glycans, including lipopolysaccharides (LPS), capsular polysaccharides, and glycoproteins, play essential roles in bacterial survival, colonization, and interaction with the host. Among the most well-known bacterial surface glycans are the lipopolysaccharides of Gram-negative bacteria. LPS is composed of lipid A, a core oligosaccharide region (commonly divided into inner and outer core), and the O-antigen, which is highly diverse between species and even strains of the same species. Gram-positive bacteria, on the other hand, are characterized by a thick peptidoglycan layer, which is often accompanied by teichoic acids or lipoteichoic acids with varying structures. Mycobacteria, the causative agents of tuberculosis and leprosy, have a distinct cell wall composition that includes peptidoglycan, arabinogalactan, and mycolic acids. This group also produces additional unique glycans such as lipomannan and trehalose-containing lipids. The incredible diversity of bacterial surface glycans contributes to their pathogenicity and ability to evade host immune responses. Understanding and characterizing these glycans are crucial for developing effective diagnostic tools, therapeutics, and vaccines against bacterial infections.

What Is a Microbial Glycan Antigen Microarray?

A microbial glycan antigen microarray (MGM) is an array of bacterial glycan antigens (often O-antigens) covalently bound to a solid support slide, against which one or many biological samples are interrogated in parallel to profile sample binding preferences and specificities. Microbial glycan antigen microarrays can contain either synthetic or isolated microbial glycans that span a wide diversity of glycan structures from known pathogens. Microbial glycans are synthesized or isolated from bacteria and derivatized and printed on solid support slides in microarray format for interrogation of a wide variety of biological samples including but not limited to antibodies, serum, whole cells, viral particles, and other glycan-binding molecules or cells.
