Sulfo-Gal

Galactose (Gal) is a common composing monosaccharide in glycoconjugates. In fact, Gal is the C4 epimer of glucose (Glc). In Gal the OH at C4 position points axially while in Glc it points equatorially. However, this slight structural modification is enough to change considerably the physicochemical and biological properties of these two hexoses.

One structural modification that improves affinity for protein interactions, especially cationic proteins, and also enhances structural specificity in molecular recognition is sulfation. Sulfation at certain positions of the Gal residues doesn’t only increase affinity for some binding proteins but also makes the structures of the controlling glycans more specific to molecular interactions. Glycans containing the sulfated Gal unit is examined across numerous multicellular organisms no matter in terrestrial or marine environments. Sulfated Gal is likely the commonest sulfated monosaccharide type. Although O-sulfation can occur at positions C2, C3, C4 or C6 of Gal, sulfation happens more often at C3 and C6 positions. The best known and likely the most abundant sulfated Gal-containing glycan in terrestrial animals is keratan sulfate (KS).

Anti-sulfo-Gal antibodies could be utilized in some areas. For instance, studies indicate that some anti-KS monoclonal antibodies (mAbs) provide useful tools to study and identify human keratinocytes. Molecules bearing carbohydrates recognized by anti-KS antibodies appear at developmentally important stages of keratinocyte differentiation, revealing that these carbohydrates may serve as markers for molecules important in the differentiation of human keratinocytes.
