Introduction to Flow Cytometry

Flow cytometry is a technique with a combination of cell counting, cell sorting, biomarker detection, and protein engineering. It is a laser- or impedance-based technology which allows simultaneous multiparametric analysis of the physical and chemical characteristics. Target particles used in flow cytometry must be suspended, and cells from solid tissue must be disaggregated before analysis. Cells or other particles are introduced into one or more laser intercept moving stream, and the resultant fluorescence or scattered laser light is captured by an appropriate detector. Flow cytometry has the ability to measure more than 10,000 particles per second. This well-established technology can be used in many areas, such as measuring the size and subcellular structures of cells, protein binding, and metabolic activities.

High Throughput Flow Cytometry

Creative Biolabs uses a novel flow cytometry-based screening platform named high throughput flow cytometry (HTFC) which incorporates the attributes of flow cytometry, HTS, and data management system into one platform. HTFC is a two-laser (red and blue), six-parameter platform with powerful data analysis capabilities. By our powerful analysis software package, this optimized HTFC system allows rapid identification of hits from multiplexed screening campaigns and can analyze up to six separate parameters over a six-decade dynamic range in each well. We offer multiplexed screening assays in 96- and 384-well plates with the speed of up to 40 wells per minute.

HTFC technology has been applied to numerous assay areas, including efflux transporter screening, in vitro toxicology, target-based screening, whole cell, and receptor binding assays.

Target-Based Screening

This assay is designed to identify hits that disrupt or augment binding to known target molecules. This assay can be illustrated by the cell-based or beads-based HCS platform. For beads-based HCS platform, each bead set can be engineered to express or capture a different target analyte, and a fluorescent reporter detected in a third parameter channel is used to quantify the analyte. Cell-based assay for target-based screening is more practical as the targets are displayed in a physiologically relevant context. Intact cell analysis for receptor-ligand interactions may minimize ligand depletion issues associated with solution-based assays.

Phenotypic Screening

The objective of cell-based assays is to identify compounds that disrupt, augment, or restore a cellular phenotype. Cell-based phenotypic screening is attractive in drug discovery as it allows interrogation of entire signaling pathway networks. Phenotypic screening is also more likely to detect undesirable or toxic effects of compounds, which impel the early stages of the drug discovery pipeline.

Fc Receptor Binding Assays

Fc-based therapeutics (e.g., antibodies and Fc-fusion proteins) mediate their effector functions through binding to Fc receptors on effector cells, thereby killing target cancer cells effectively. Consequently, measuring the binding of antibody candidates to specific Fc receptors is a critical step to assess antibody efficacy and safety. Different methods have been used for this purpose, among which the flow cytometry is often used due to its high levels of accuracy, precision, and linearity, making it suitable for comparability, potency, and stability assays. Moreover, it can be readily used to detect structural differences such as glycosylation, which affect Fc receptor binding.
