Introduction of Air Pouch Model
The air pouch is developed simply by subcutaneous (s.c.) injection of sterile air into the intra-scapular area of the back of a rat or mouse. This procedure induces the proliferation of cells that stratify on the surface of the cavity to form a cellular lining consisting of macrophages and fibroblasts, which is morphologically similar to that found in synovial cavity. Later, local inflammation can be induced by injecting with carrageenan solution or other irritants (e.g. LPS, zymosan or bradykinin) into the air pouch. Here, the pouch serves as a reservoir of locally accumulated cells and mediators that can be easily measured in the fluid. These endpoints can be quantified and used to determine the degree of inflammation, resolution of inflammation, or anti-inflammatory activity of drugs.
Applications of Air Pouch Model
The subcutaneous air pouch is an in vivo model used to study acute/chronic inflammation, granulomatous inflammation, the resolution of the acute inflammatory response, and evaluation of oxidative stress. In preclinical studies, it is frequently used to screen potential anti-inflammatory compounds.
Advantages of Air Pouch Model
There are several advantages of the air pouch over the injection of irritants directly into the knee joint of animals: Firstly, the volume of exudate produced in the air pouch is much larger, enabling the measurement of multiple parameters from the same animal. Secondly, air pouch model is much easier to manipulate because it is technically challenging to perform procedures including direct injection of irritants to a knee joint and subsequent collection of inflammatory exudates without causing damage leading to inflammation.
