The Tol2 transposon element was originally identified in the genome of the medaka fish and has now been extensively used to make transgenic zebrafish model for various studies. Afterward, this Tol2 transposon was discovered to be active in many other vertebrates, including zebrafish, chicken, mouse, human and so on. However, as an alternative way of DNA microinjection, Tol2 has not yet been widely used in mouse transgenesis. Types of Tol2 Transposon

Generally, wild-type transposons exist as both ‘autonomous' and ‘non-autonomous' versions. The Tol2 element is an autonomously active transposon, containing a gene encoding a complete and functional transposase that is capable of identifying, excising, and reinserting the DNA element defined by its inverted terminal repeats (ITR) or other elements with the same ITRs. Non-autonomous elements, which are no longer able to produce their own transposase protein due to mutations or deletions of the coding region of the transposase, can also be catalyzed by this functional enzyme.

Features of Tol2 Transposon System

  • The integration of the targeted gene by Tol2 is mediated by a ‘cut-and-paste' mechanism, which does not cause any rearrangement or modification at the target site except for the creation of an 8-base pair (bp) duplication.
  • Tol2 transposon shows no preferences with respect to the integration sites which means theoretically it can translocate to anywhere on a chromosome.
  • Single copy insertions can be achieved and DNA inserts of fairly large sizes (as large as 13 kilobases) can be cloned between these sequences. This is a considerable advantage compared to Tc1/mariner-type transposons such as Sleeping Beauty, which shows a cargo size limitation.
  • Transgenes cloned on this Tol2 vector are reliably expressed in both transgenic animals and cells, even with larger fragment insertions.

Applications of Tol2 Transposon System

Due to the high efficiency and abilities to carry larger foreign DNA fragments, Tol2 transposon system is discovered to be useful in not only the establishment of stable expression cell strain but also the generation of transgenic mice and rats. By injecting a foreign DNA cloned into a Tol2-transposon vector together with the transposase mRNA into the cytoplasm of fertilized eggs, we can obtain the Founder mice simply and easily.
