Syngeneic mice-derived models could recapitulate the original disease significantly better than traditional in vitro immortalized cell line derived syngeneic models. Syngeneic Mice-derived Tumor Models
Syngeneic mice-derived models are homografts of spontaneous murine tumors, which could be regarded as “a mouse version of PDX (Patient-Derived Xenograft)”, are primary tumors in nature and never manipulated in vitro, being capable of reflecting original mouse genetic profiles and tumor histopathology.
To some degree, syngeneic mice-derived models recapitulate the original disease significantly better than traditional in vitro immortalized cell line derived syngeneic models. They are cancer stem cells (CSCs) driven diseases, which maintain heterogeneous histopathology, rich microenvironments and different differentiation phenotypes.
Features and Applications
Since they are never cultured in vitro, syngeneic mice-derived tumors show quite a contrast to cell line based-syngeneic models in cytobiological characteristics: difficult to culture in vitro, requiring special hormones or growth factors which is relevant to massive apoptosis growth arrest even for short-term culture.
Similar to PDX, syngeneic mice-derived tumor models are expected to enable the study of predictive biomarkers for targeted therapeutics and offers advanced predictive power as preclinical cancer models.
Any kind of spontaneous mouse tumors can be used as the sources of syngeneic mice-derived tumor models, including:
a. spontaneous tumors from GEMMs
b. spontaneous tumors due to aging
c. tumors from those induced by chemical carcinogens
