Subcutaneous Tumor Model

Animal models have been widely used as the front line in predicting efficacy and finding toxicities for cancer chemotherapeutic agents before entering the clinic. This has resulted in the development of many different types of animal models of human cancer. Tumor models are demonstrated being useful in identifying novel compounds that subsequently demonstrate significant clinical activity against the appropriate histology. Subcutaneous tumor model is the most popular assessment system for in vivo efficacy evaluation of novel anti-cancer medicine candidates. In general operations, tumor cells are usually subcutaneously implanted on the flank or dorsum of animals (mostly mice or rats), in which the transplanted cells proliferate and form measurable tumors.

Subcutaneous tumor implants are more arduous since the candidate agent have to cross physiologic barriers to reach the target tumor. Furthermore, the subcutaneous models require higher requirements for monitoring and evaluation.

Subcutaneously transplanted tumor cells are proved to be a convenient approach to evaluate novel potential anti-cancer drugs in vivo. A wide variety of human and murine cell lines derived from both, including solid tumors and leukemias, while covering a comprehensive gene phenotypes of the tumor, have been adapted to grow in a rodent host and thus allow detection of compounds in the relevant tumor model.
