Importance of ADA Quantification Assays
During the past years, monoclonal antibody has greatly promoted the development of drug discovery. Even though with highly tolerability and effectiveness, humanization mAbs can still be immunogenic, leading to the development of ADA. Especially, high-titer and high-affinity ADA could cause impaired therapeutic efficacy or adverse events. Therefore, accurate and reliable detection of ADA is therefore necessary. Recently, several assay formats have been used to detect ADA concentrations.

ADA Quantification Assays Provided by Creative Biolabs
In the past, two major problems have hampered the development of ADA quantification. An important challenge is the presence of mAb itself in the samples, which complicates the ADA measurements and results in an underestimation of ADA. Another issue is the large heterogeneity in reported results, which are dependent on the standard, the assay type, dilution of the sample, and the lack of universal standards for ADA assays. To solve these matters, drug-tolerant assays have been developed to measure ADA in complex (using drug/ADA complex precipitation in combination with acidification).
