Immunodeficient mice engrafted with human cells and tissues have provided a promising alternative to in vitro studies with human tissues for the study of human immunobiology. The immunodeficient mice can permit efficient engraftment with human hematopoietic cells and then generate a functional human immune system. These humanized mouse models are becoming increasingly important for preclinical studies of human immunity, tissue regeneration, hematopoiesis, cancer, and infectious diseases etc. One approach for generating humanized mice to investigate the human immune system is to engraft mature lymphoid cells, which is so-called the human peripheral blood lymphocyte SCID (Hu-PBL- SCID) model. Humanized PBL SCID Mouse

The severe combined immunodeficiency (SCID) is a severe immunodeficiency genetic disorder that is characterized by the complete disability of the adaptive immune system to mount, coordinate, and sustain an appropriate immune response, generally due to the absence of T and B lymphocytes. Severe combined immunodeficiency (SClD) mice can be stably engrafted with human peripheral blood lymphocytes, generating humanized PBL SCID chimeras; basically, these are mice with a human immune system.

The Hu-PBL-SCID model is established simply by injection of human peripheral blood mononuclear cells or spleen or lymph node cells into immunodeficient mice. Recent modifications of the humanized PBL-SCID mouse models have allowed investigation of the infiltration and rejection of human islet, skin and arterial allografts and the cell populations related to the immune rejection process.
