Sleeping Beauty (SB) transposon is a synthetic transposon of the Tc1-like mariner family. It was originally discovered in salmon, where it had been silenced through evolution and was then reconstructed by investigators based on molecular phylogenetic data to recover its ability to transpose the genome. The first functional version of SB transposon is SB10, which was so named because the reconstruction process required ten major steps. Thereafter, continued efforts have been made by researchers in order to improve its ability of gene transfer. As a result, a novel SB transposase, SB100X, which shows high efficiency of stable gene transfer comparable to stable transduction efficiencies of integrating viral vectors, was developed to greatly boost its research application.

The Sleeping Beauty System

The Sleeping Beauty transposon is a two-component system comprised of an SB transposase and a transposon. The transposase is the enzyme required for catalysis of transposition and the transposon contains an expression cassette of interest which is flanked by two inverted repeat/direct repeat (IR/DR) sequences. By specifically binding to these sequences, transposase mediates the excision and precise integration of the target cassette into the specific sites of the genome. Generally, the specific integration site for SB transposon is the TA dinucleotide base pair.

Features of Sleeping Beauty Transposon System

Compared to other methods of gene transfer such as the use of viral vectors and naked DNA, SB transposon exhibits great advantages. For viral vectors, although high efficiency can be obtained, they can be susceptible to cells' defense mechanisms. Besides, they are more efficient than plasmid-based methods. In summary, transposons combine the advantages of these two methods and avoid many of these problems.

When compared to other transposons, however, SB shows lower tolerance of cargo size, which means increased cargo size exponentially decreases the efficiency of transposition. As is reported, when the size of the payload between IR/DRs increases above 2 kb, the transposition efficiency drops dramatically. But still, SB transposon system is widely used in different fields and modified versions have been developed to expand its application.
