Antibody Isoform Analysis

Antibodies exist in various isoforms and the various isoforms differ in their biological features, structure, target specificity and distribution. The complex and lengthy upstream and downstream production methods of the antibodies make them susceptible to physical and chemical modifications.   • Capillary isoelectric focusing (CEIF)
  • Matrix-assisted Laser Desorption/Ionization (MALDI)

MALDI is a technique for soft ionization of mass spectrometry (MS) that uses a laser energy absorbing matrix to create ions from large molecules with minimal fragmentation. For instance, the MALDI analysis of the IgG1 product indicates that most of the N-linked glycans were complex, biantennary glycans with core fucosylation. Small amounts of triantennary and high-man-nose glycans were also observed. A significant proportion of the N-glycans contain terminal α-gal and N-glycolyl neuraminic acid (NGNA) residues

  • Electrospray Ionization Mass Spectrometry (ESI-MS)

Multiple charge isoforms of mAb could be separated by cation-exchange chromatography, and their identities will be established primarily by ESI-MS analysis of the intact, reduced, and deglycosylated molecule.
