Application NotesThe kit is a fluorescence-based assay that utilizes the preferred cathepsin-B substrate sequence RR labeled with AFC (amino-4-trifluoromethyl coumarin).
Molecular Mechanism of ActionUpon incubation with the cathepsin-B enzyme, the synthetic substrate RR-AFC is cleaved, resulting in the release of free AFC. The amount of AFC can be measured using a fluorometer or fluorescence plate reader at an excitation wavelength of 400 nm and an emission wavelength of 505 nm. This allows for quantification of cathepsin-B activity in cell lysates or other samples.
