| Target/Description | Enzymes known as isocitrate dehydrogenases facilitate the oxidative decarboxylation of isocitrate to 2-oxoglutarate. Mutations in isocitrate dehydrogenase 1 and 2 (IDH1 and IDH2) have been observed in various forms of human cancer. In contrast to the wild-type IDH1, which generates α-ketoglutarate (α-KG) by reducing NADP⁺ to NADPH, the IDH1(R132H) mutant facilitates the conversion of α-ketoglutarate to 2-hydroxyglutarate (2-HG) by oxidizing NADPH to NADP⁺. |
