| Target/Description | This kit utilizes the sandwich enzyme-linked immune-sorbent assay (ELISA) technology for detection. The capture antibody is precoated on a 96-well plate, with the biotin-conjugated antibody used for detection. Standards and samples are added to the wells and allowed to incubate. Unbound conjugates are then removed with wash buffer. Biotinylated detection antibody is added to bind with the target, followed by the addition of HRP-Streptavidin. After a final wash, TMB substrates are added to visualize the HRP enzymatic reaction. The TMB substrate is catalyzed by HRP to produce a blue product, which turns yellow with the addition of an acidic stop solution. The optical density (O.D.) absorbance is read at 450nm with a microplate reader. The concentration of the target in the sample correlates positively with the O.D. reading and can be calculated by comparing it with the standard curve. |
