| Target/Description | The assay involves transfecting cells with a CRE luciferase reporter. CRE reporter contains the firefly luciferase gene controlled by cAMP response element (CRE). Elevation of cAMP activates CREB to bind CRE, leading to luciferase expression. Forskolin boosts cAMP levels, inducing luciferase expression. Co-transfection of PDE4D7 reduces forskolin-induced cAMP and lowers luciferase levels. Treatment with a PDE4D inhibitor restores cAMP levels and increases luciferase activity. |
