Introduction of MALS

MALS is a well-established technique for measuring the size and size distribution of molecules and particles dispersed in a liquid. The principle of this technique is to analyze the temporal fluctuations of light scattered by the particles at a given scattering angle and the particle size is calculated by using the Stokes-Einstein relationship. Compared to single-angle dynamic light scattering, MALS provides more robust, reproducible and accurate particle size distributions. This technique is also used to directly measure molar mass of macromolecules and complexes in solution. By utilizing MALS in conjunction with size exclusion chromatography (SEC) and an on-line concentration detector, the absolute molar mass and size of biological macromolecules can be determined. Furthermore, composition-gradient MALS (CG-MALS) uses a composition-gradient system coupled to MALS/RI/UV detectors to determine interactions between biological macromolecules including protein-protein or protein-DNA complexes, without the need for labeling or immobilization, by directly detect changes of molar mass due to complex formation or dissociation in solution.
